David Tereshchuk: Celebrating a Virtually Forgotten Media Maestro

December's days are to dwindle down to year's-end, and I'm already into holiday time's mental excursions. They're taking me off-road a bit, into the byways of no-longer-mainstream media.

Staffers at the University of California Press have claimed over the past year or so some surprise at the success enjoyed by their publication (finally allowed by the author's own jokey edict suppressing it for 100 years) of Mark Twain's Autobiography. And the three-volume tome -- unexpurgated, but written cagily, and yet still self-revealingly at times -- has prompted me to pursue one of the great humorist's own personal delights.

Twain in fact delighted in another humorist, one definitely worthy of his admiration, and a man who shared with him membership of the mordantly named "Damned Human Race Luncheon Club" -- a gathering noted as much for cigars and wise-cracking cynicism as it was for any eating of lunch.

I speak of the virtually forgotten columnist, Finley Peter Dunne, who wrote in a range of Chicago newspapers - notably the now-dead Post and the now severely injured Tribune. Much of the time he spoke through the invented personage of "Martin J. Dooley," a down-to-earth Irish bartender whose wisdom, Mr. Dooley's Opinions, began to appear collected in book form at the turn of the 20th century.

Often enough this sardonic expert on conviviality didn't range beyond the bar room -- vouchsafing for example, and presumably from experience, that "alcohol is necessary for a man, so that now and then he can have a good opinion of himself, undisturbed by the facts."

More broadly, though, and for students of the media especially, Dunne should be remembered at the least for being the first to proclaim (though who now remembers it was Dunne?) that one role of the press is to "comfort the afflicted, and afflict the comfortable."

He has long been cheated out of credit, too, for another famous adage. The redoubtable 1980s speaker of the U.S. House of Representatives, Tip O'Neill, has too readily been quoted as the author of "All politics is local." In fact it was Dunne, or at least his mouthpiece behind the bar, who said that first.

Another "Mr. Dooley" one-liner originally appeared to be a comment on late 19th century events, but nowadays I feel it could be a contender for quotation in perpetuity -- "The Democratic Party ain't on speaking terms with itself."

In today's climate of personally vituperative political discourse -- often as small-minded as it is intemperate -- it's worthwhile recalling a very different kind of exchange between Dunne and a leading politician of that earlier time.

When Theodore Roosevelt published his Rough Riders book, to celebrate his (or more properly his small regiment's) role in the Spanish American War, Dunne gave it a jocular review, highlighting its self-serving qualities by saying -- infamously -- that it should have been titled Alone in Cuba!

Riots of hilarity greeted that single pointed quip in the salons of Washington and the smoking-rooms of the entire nation. Roosevelt himself took it in good part, writing to Dunne that "I regret to state that my family and intimate friends are delighted with your review of my book. Now I think you owe me one; and I shall expect that when you next come East you pay me a visit."

The two men did meet, in 1900, and became good friends. Dunne, like the best columnists, remained a sharp reporter as well as an opinionator -- and scooped all his colleagues with the news (from a most reliable source, the horse's mouth) that Roosevelt was going to run as vice president to William McKinley.

Dunne's special scorn was often reserved for those individuals who are intolerably intolerant. And I wonder -- if he were alive now, amid all today's dogmatism and bigotry -- whether he might like to revisit the Mr. Dooley aphorism that most caught my attention at this celebratory season: "Thanksgiving was founded by the Puritans to give thanks for being preserved from the Indians, and we keep it to give thanks we are preserved from the Puritans."

By msnbc.com news services

Wall Street rallied at Friday?s opening bell, as investors were boosted by an upbeat report on the U.S. labor market.

The Dow Jones industrial average was lately up?about?120 points.

Employment growth picked up speed in November, pushing down the nation?s unemployment rate to 8.6 percent, the Labor Department reported Friday.

Nonfarm U.S. payrolls increased by 120,000 jobs last month, outpacing October?s 80,000 gain and roughly matching analysts? expectations. The drop in the jobless rate fell to its lowest level since March 2009, down from the prior month?s 9 percent level.

Investors were also focused on a summit that could decide the euro zone's future.

Signs that leaders of France and Germany are working hard to reach a compromise deal ahead of the Dec. 9 summit, viewed as make-or-break for the 12-year old single currency bloc, kept the euro firm versus the dollar and encouraged investors to push equity markets higher.

German Chancellor Angela Merkel vowed in a speech to defend the euro, though she warned the euro zone debt crisis would take years to resolve.

Data on Thursday showed U.S. manufacturing activity rose to its highest in five months, while consumer spending and private-sector job creation have also been stronger.

The Associated Press and Reuters contributed to this report.

Reports: Myanmar's Shan rebels sign cease-fire (AP)

BANGKOK ? One of the main ethnic rebel groups battling Myanmar's government signed a preliminary cease-fire Friday, websites operated by exiled journalists reported.

The reported agreement comes as Myanmar's army-backed but elected government is seeking international legitimacy through democratic reforms after years of military repression. U.S. Secretary of State Hillary Rodham Clinton just concluded a visit to the country that was intended to encourage those reforms.

The Shan State Army-South rebel group was one of the biggest not to previously sign a cease-fire with the government.

The Thailand-based Irrawaddy website reported the rebels signed an agreement with Myanmar's official Shan State government. India-based Mizzima News and the Shan Herald Agency for News, a website close to the guerrillas, reported similar news.

During her visit that ended Friday, Clinton offered modest incentives to the new government, while calling on it to end brutal campaigns against ethnic minorities, free all political prisoners and break military ties with North Korea.

Myanmar for decades has been at odds with the ethnic groups who seek greater autonomy, but a military junta that took power in 1988 signed cease-fire agreements with many. Some of those pacts have been strained as the central government sought to consolidate power, and combat resumed.

Neither the government nor the rebel group would immediately confirm the new cease-fire, but the Shan Herald Agency's report cited Shan rebel leader Lt. Gen. Yawdserk saying an agreement was reached on a cease-fire, political negotiations, development and cooperation against drugs.

It was not clear when or if the Shan group would sign a cease-fire agreement with the central government.

Myanmar's government in recent weeks has held high-level but low-profile talks with rebel groups with which it has never signed cease-fires, or had cease-fires that have broken down. The groups reportedly involved in talks include the Shan, Karen, Karenni, Chin and Kachin.

A high-level government delegation met Tuesday with the Kachin Independence Organization in Ruili in China's Yunnan province, Myanmar's state press reported this week. The Kachin, whose state is in the north, have been fighting the government since June, when the army tried to break up some of their militia strongholds.

The reports said both sides agreed at the meeting to continue the initial peace talks aimed at a cease-fire and political dialogues.

Kachin sympathizers have circulated accounts of government brutality, but the remote area is mostly inaccessible to foreigners and the allegations are difficult to confirm. The government has reported little on the fighting.

Myanmar democracy leader Aung San Suu Kyi has described an end to the fighting with ethnic guerrillas as national priority, and last month said she would be willing to help with peace negotiations.

Pre-installed Android software poses massive security risk (Appolicious)

Android?s seen its share of privacy issues and data safety concerns, and while frequent malware attacks on the platform have spurned an entire industry, every week seems to bring news of even more dire straits for the Android camp. Carrier IQ is the latest to come under fire, after Android security researcher Trevor Eckhart showed how the mobile device tracker logs each keystroke, sending the data to unknown locations. Carrier IQ?s software is used by carriers and companies to obtain information on end user behavior, with the intent of improving the customer experience and to better understand device issues. But Eckhart?s expos?, which includes a detailed 17-minute video, has shed new light on the methods behind Carrier IQ, leaving many to wonder the true objectives behind software that?s sending private data to unverified places.

Things got ugly when Eckhart called Carrier IQ?s software a ?rootkit? because of its ability to access device data while concealing its presence. This isn?t an app you download from the Market, but software pre-installed in your device. Carrier IQ sent Eckhart a cease-and-desist letter, demanding an apology for calling the software a rootkit, but later sent out an apology of their own after the Electronic Frontier Foundation deemed Eckhart?s comments and research as protected under the Copyright Act?s fair use provision. But while Eckhart?s stuck in a song and dance routine with Carrier IQ, consumers are still wondering why their Android phones have software that logs keystrokes, phone numbers, and even SMS content.

Pre-installed apps are security risks

Another team of researchers at North Carolina State University have given consumers even more reason to worry, having uncovered a number of vulnerabilities in the standard configurations of popular Android devices from the likes of Motorola, HTC and even Samsung. In a paper published by Michael Grace, Yajin Zhou, Zhi Want and Xuxian Jiang, the team demonstrates how these vulnerabilities could be used by an untrusted app to send SMS messages, record conversations or wipe all user data from the handset without needing user permission.

The HTC Legend, EVO 4G and Wildfire S were just some of the devices tested by the NC University team, unveiling a slew of new security leaks, even from pre-loaded applications that not only come standard on many Android handsets, but can?t be easily uninstalled.

Between Eckhart?s report and the paper from NC University, it appears our Android phones come full of security holes right out the box. What will Google do about its OEM partners that are sending out devices with such high potential for attackers, made especially attractive because of their massive install base?

New 'Achilles' heel' in breast cancer: Tumor cell mitochondria

Thursday, December 1, 2011

Researchers at the Kimmel Cancer Center at Jefferson have identified cancer cell mitochondria as the unsuspecting powerhouse and "Achilles' heel" of tumor growth, opening up the door for new therapeutic targets in breast cancer and other tumor types.

Reporting in the online Dec.1 issue of Cell Cycle, Michael P. Lisanti, M.D., Ph.D., Professor and Chair of Stem Cell Biology & Regenerative Medicine at Thomas Jefferson University, and colleagues provide the first in vivo evidence that breast cancer cells perform enhanced mitochondrial oxidative phosphorylation (OXPHOS) to produce high amounts of energy.

"We and others have now shown that cancer is a 'parasitic disease' that steals energy from the host -- your body," Dr. Lisanti said, "but this is the first time we've shown in human breast tissue that cancer cell mitochondria are calling the shots and could ultimately be manipulated in our favor."

Mitochondria are the energy-producing power-plants in normal cells. However, cancer cells have amplified this energy-producing mechanism, with at least five times as much energy-producing capacity, compared with normal cells. Simply put, mitochondria are the powerhouse of cancer cells and they fuel tumor growth and metastasis.

The research presented in the study further supports the idea that blocking this activity with a mitochondrial inhibitor -- for instance, an off-patent generic drug used to treat diabetes known as Metformin -- can reverse tumor growth and chemotherapy resistance. This new concept could radically change how we treat cancer patients, and stimulate new metabolic strategies for cancer prevention and therapy.

Investigating the Powerhouse

Whether cancer cells have functional mitochondria has been a hotly debated topic for the past 85 years. It was argued that cancer cells don't use mitochondria, but instead use glycolysis exclusively; this is known as the Warburg Effect. But researchers at the Jefferson's KCC have shown that this inefficient method of producing energy actually takes place in the surrounding host stromal cells, rather then in epithelial cancer cells. This process then provides abundant mitochondrial fuel for cancer cells. They've coined this the "Reverse Warburg Effect," the opposite or reverse of the existing paradigm.

To study mitochondria's role directly, the researchers, including co-author and collaborator Federica Sotgia, Assistant Professor in the Department of Cancer Biology, looked at mitochondrial function using COX activity staining in human breast cancer samples. Previously, this simple stain was only applied to muscle tissue, a mitochondrial-rich tissue.

Researchers found that human breast cancer epithelial cells showed amplified levels of mitochondrial activity. In contrast, adjacent stromal tissues showed little or no mitochondrial oxidative capacity, consistent with the new paradigm. These findings were further validated using a computer-based informatics approach with gene profiles from over 2,000 human breast cancer samples.

It is now clear that cancer cell mitochondria play a key role in "parasitic" energy transfer between normal fibroblasts and cancer cells, fueling tumor growth and metastasis.

"We have presented new evidence that cancer cell mitochondria are at the heart of tumor cell growth and metastasis," Dr. Lisanti said. "Metabolically, the drug Metformin prevents cancer cells from using their mitochondria, induces glycolysis and lactate production, and shifts cancer cells toward the conventional 'Warburg Effect'. This effectively starves the cancer cells to death".

Personalized Treatment

Although COX mitochondrial activity staining had never been applied to cancer tissues, it could now be used routinely to distinguish cancer cells from normal cells, and to establish negative margins during cancer surgery. And this is a very cost-effective test, since it has been used routinely for muscle-tissue for over 50 years, but not for cancer diagnosis.

What's more, it appears that upregulation of mitochondrial activity is a common feature of human breast cancer cells, and is associated with both estrogen receptor positive (ER+) and negative (ER-) disease. Outcome analysis indicated that this mitochondrial gene signature is also associated with an increased risk of tumor cell metastasis, particularly in ER-negative (ER-) patients.

"Mitochondria are the 'Achilles' heel' of tumor cells," Dr. Lisanti said. "And we believe that targeting mitochondrial metabolism has broad implications for both cancer diagnostics and therapeutics, and could be exploited in the pursuit of personalized cancer medicine."

Samsung's mSATA PM830 is eight grams of pure SSD

The newest member of Samsung's 6Gb/s PM830 family of SSDs is designed to sit inside the Ultrabooks of 2012, which means it's had go through some slimming down. The mSATA drive uses 20 nanometer NAND for face-melting speed, but is crammed into a 50.95 x 30 x 3.8mm package, weighing only eight grams. The drive will comfortably read 500MB/s and write 260MB/s on a clear day, six times faster than a comparable HDD. It'll allow Windows to boot in under ten seconds and transfer "five DVD files" (roughly 45GB) in around a minute. The drives also come with 256-bit AES for those moments when your Ultrabook gets stolen, or more likely, floats away in the wind. 64GB, 128GB and 256GB variants will roll out to OEMs shortly but we expect a consumer version to be announced shortly for everyone with some courage and a screwdriver.
